Hot Start PCR

  • Hot Start PCR prevents misextension from non-specifically annealed primers and primer-dimers.
  • HOT FIREPol®  DNA Polymerase is a chemically modified FIREPol®  DNA Polymerase. At ambient temperature it is inactive having no polymerization activity. HOT FIREPol® DNA Polymerase is activated by a 12-15 min incubation step at 95º C.
  • Hot-start master mixes are also available in ready-to-load format with variety of MgCl2 concentrations.
  • 5x HOT FIREPol® Blend Master Mix is a highly user-friendly and versatile tool for your Hot Start PCR. The mix is based on a blend of our HOT FIREPol®polymerase and a proofreading polymerase, providing a low error rate and high sensitivity. In addition, the mix also allows amplification of longer fragments (up to 5 kb). Here some great advantages of this ready-to-load mix: decreased set-up time
    • reduced risk of contamination
    • less pipetting mistakes
    • consistent reaction to reaction performance
    • contains all reagents required for PCR
  • Like all products of Solis BioDyne, the 5x HOT FIREPol® Blend Master Mix is stable at ambient temperature for up to one month. This makes handling and set-up even more convenient, since no ice has to be used.

PCR

  • The polymerase chain reaction (PCR) is a technique to amplify a short sequence of DNA even in samples containing only minute quantities of DNA. PCR is used to amplify selected sections of DNA or RNA across several orders of magnitude.
  • FIREPol® is highly processive, thermostable and robust Taq polymerase with amplification range up to 3 kb. Due to Solis Tag technology, FIREPol® has an enhanced stability at ambient temperature with no activity loss up to 1 month.
  • We supply FIREPol® Polymerase as enzyme kit with a selection of buffers and an enhancer for personal optimization.
  • Master Mixes with or without ready-to-load feature are also available.

High Resolution Melt Analysis (HRM)

  • High Resolution Melting (HRM) is a hugely powerful, close-tube, post-qPCR genotyping method for the detection of mutations, polymorphisms and epigenetic differences in double stranded DNA samples.
  • It goes beyond the power of classical melting curve analysis (MCA) by allowing to study the thermal denaturation of a double-stranded DNA in much more detail. Samples can be discriminated according to their sequence, length, GC content or strand complementarity. Even single base changes such as SNPs (single nucleotide polymorphisms) can be readily identified.
  • HOT FIREPol® EvaGreen® HRM Mix is an optimized ready-to-use solution for High Resolution Melt (HRM) Analysis, incorporating EvaGreen® dye.
  • It includes:  HOT FIREPol® DNA Polymerase, ultrapure dNTPs, MgCl2 and ROX dye according to system requirements.
